A quantum dot fluorescent microsphere based immunochromatographic strip for detection of brucellosis

Abstract Background Brucellosis is a serious zoonosis disease that frequently causes significant economic loss in animal husbandry and threatens human health. Therefore, we established rapid, accurate, simple sensitive fluorescent immunochromatographic strip test (ICST) based on quantum dots (QDs) for detection the antibodies of Brucella infection animals serum. Results The strips were successfully prepared by dot microspheres (QDFM) as tracers, which covalently coupled to an outer membrane protein OMP22. OMP28 monoclonal OMP22 separately dispensed onto nitrocellulose quality control lines, respectively. critical threshold determining negative or positive through ratio signal line (H T / H C ) 0.0492. repeatability was excellent with overall average CV 8.78%. Under optimum conditions, limit 1.05 ng/mL (1:512 dilution). With regard brucellosis 150 clinical samples, total coincidence rate ICST Rose Bengal plate (RBPT) 97.3%, samples 98.8%, 95.3%, sensitivity RBPT 1:32, no cross reaction sera other related diseases observed. Conclusion In our present study, QDFM has promising application on-site screening owing its high speed, sensitivity, specificity low cost.
